Postdoctoral researcher in T cell engineering and immuno-oncology

To strengthen the laboratory of Professor Caroline Arber (https://www.unil.ch/dof/arber-barth), we are looking for a Post-Doctoral Research Fellow in T cell engineering.

Afin de renforcer le laboratoire de recherche en oncologie (LRO) de Mme la Pre Caroline Arber (https://www.unil.ch/dof/arber-barth) nous recherchons un-e Post-Doctorant-e dans le domaine de l’ingénierie de lymphocytes T.

Contexte

The Lausanne University Hospital (CHUV) is one of five Swiss university hospitals. Through its collaboration with the Faculty of Biology and Medicine of the University of Lausanne and the EPFL, CHUV plays a leading role in the areas of medical care, medical research and training.

The Department of Oncology UNIL-CHUV brings together clinical and cancer research in a strong spirit of innovation. It comprises the services of medical oncology, immuno-oncology including advanced cellular therapies, radiation oncology, hematology including stem cell transplantation and the center for experimental therapies. It also integrates the Lausanne branch of the Ludwig Institute for Cancer Research (LICR), particularly in the field of immunotherapy.

Notre département d’oncologie UNIL-CHUV réunit la clinique et la recherche sur le cancer dans un esprit d'innovation. Il regroupe les services d'oncologie médicale, d’immuno-oncologie, de radio-oncologie, d'hématologie avec l'activité de transplantation de la moelle osseuse du CHUV et le centre de thérapies expérimentales. Il intègre également la branche lausannoise du Ludwig Institute for Cancer Research (LICR) pour la recherche sur le cancer, en particulier dans le domaine de l'immunothérapie.

Mission

As part of your mission, you will be will be working on a project developing allogeneic off-the-shelf chimeric antigen receptor (CAR) T cells that resist T and NK cell mediated rejection in allogeneic hosts. Thus, you will:

• Generate and characterise in vitro the newly designed off-the-shelf allogeneic CAR-T cells targeted against acute myeloid leukaemia (AML). These CAR-T cells are engineered to resist rejection by T and NK cells of allogeneic hosts. You will perform the immunological and functional characterization of this new type of CAR-T cell product (test their capacity to resist or evade immune rejection, asses their anti-leukemic activity)
• Perform animal experimentation in accordance with the existing authorizations
• Assess the in vivo function of the newly designed off-the-shelf allogeneic CAR-T cells, by analysing their anti-leukemic activity in vivo in mouse xenograft models (monitoring of tumor growth by bioluminescent imaging and other techniques), evaluate their capacity to resist T and NK cell mediated rejection in vivo
• Participate actively to the oral presentations of the group, within the department, and in national and international conferences
• Help preparing scientific reports and new funding request for the laboratory (in English)
• Assist the team leader with the supervision of students (Master’s and PhD) and laboratory technicians.

Dans le cadre de la mission de développer une nouvelle thérapie cellulaire à base de lymphocytes T allogéniques exprimant un récépteur antigénique chimérique (CAR), vous serez amené-e à étudier leur capacité de résister à la réjection par les lymphocytes T et NK de l’hôte et effectuer leur fonction anti-leucémique :

• Générer et caractériser in vitro des lymphocytes CAR-T allogéniques ‘off-the-shelf’, ciblé contre la leucémie myeloïde aiguë, qui résistent à la rejection des cellules T et NK de l’hôte allogénique (caractérisation immunologique, capacité de supprimer ou évader à la réaction de rejection, fonction anti-leucémique)
• Effectuer des experimentations animalières en conformité avec les autorisations
• Analyser in vivo la fonction des lymphocytes CAR-T allogéniques ‘off-the-shelf’: analyser la fonction anti-leucémique des lymphocytes CAR-T (monitorer la croissance des tumeurs dans les souris par imagerie bioluminescente et d’autres techniques), évaluer la résistance des lymphocytes CAR-T à la réjection par des cellules T et NK allogéniques
• Participer activement aux présentations orales du groupe, dans le département et dans des conférences nationales ou internationales (en anglais)
• Assister le team leader dans la supervision des étudiant-e-s et des technicien-ne-s de laboratoire.

Profil

• You have a PhD or equivalent in immunology or cancer biology or in immunooncology
• You have experience with mouse models (LTK1 certificate or equivalent is required) and direct hands-on experience of at least 1 year with mouse models
• Experience in human immunology and human lymphocyte engineering represents a strong asset
• You have a strong knowledge in flow cytometry and in animal experimentation
• You have a good command of the usual computer tools
• You are fluent in English, both written and spoken, including for scientific presentations
• You are dynamic, adaptable and flexible.
• Vous êtes au bénéfice d'un PhD ou d'un titre jugé équivalent en immunologie, en biologie du cancer ou en immunooncologie
• Vous justifiez d'une expérience avec des modèles de souris (minimum certificat LTK1 ou équivalent) ainsi que d'une expérience d'au moins 1 an avec des modèles souris de cancer
• Expérience en immunologie humaine et en ingénierie des lymphocytes humains représente un réel atout.
• Vous démontrez de très bonnes compétences en analyses de cytométrie de flux (FACS) ainsi qu'en expérimentation animalière
• Vous avez une bonne maîtrise des outils informatiques usuels
• Vous êtes à l’aise en anglais à l’oral comme à l’écrit, inclus pour des présentations scientifiques
• Dynamique, vous faites preuve d’adaptation et de flexibilité.
